You can, but it's kind of hard to get an accurate reading that way, since you're looking at it from an angle.

Most take a sample with a thief or turkey baster or something and take a reading in that...they sell little tubes on a stand for that purpose.

They're called hydrometer test jars on Northern Brewer.
 
you can also use the plastic tube that the hydrometer comes in. i filled mine up with enough water to where i was sure i could get a floating hydrometer, then marked it with a black magic marker. so now i just use my turkey baster and fill the tube up to the mark and im "good to go" ( i hate the new taco bell commercials.)
