I attempted to harvest yeast from the bottom of my carboy for the first time yesterday, however I may have made a very stupid mistake.

After I racked the beer to a secondary, I used pre-boiled water to slosh around and loosen up the yeast cake. However, I did not let this water cool down to a reasonable temperature - I didn't take a temp reading but my guess was that it was at least 150F.

Thing is, it was probably only a cup or a cup and a half, whereas there was probably a solid 3 or 4 cups worth of yeast in the carboy. Maybe I'm trying to be too optimistic but maybe it cooled down when it was mixed in :drunk:

Do you think my mistake killed the yeast, rendering it unusuable? Is there any harm in attempting to pitch this yeast next time and if it doesn't start fermenting within a day or two, add a pouch of dry yeast to pick it back up?

You probally killed some yeast when it first hit, but cooled down as it mixed with what was in there and the rest of the yeast survived. You want to make a starter anyway, so by doing so you will be able to determine if any yeast survived. Not only just determine that some survived, but to increase the amount of living yeast cells.
