So i washed my yeast cake and got about 3/4 gallon of trub/yeast/ water mixture in a boiled-sterilized 1 gallon plastic milk carton. Waiting for it to settle out. Should I leave it at room temp over night or put in the fridge?

You want to let the yeast slurry sit for about 20 minutes, room temperature is fine. Leaving the bottom 1/2" layer of trub in the bottom of the gallon jug, pour the milky liquid into your sanitized permanent storage vessel. Store the now-washed yeast in the bottom/back of your fridge.
