Yes. Extreme over pitching can result in unwanted flavors in your beer.
Using the proper amount isn't really an exact science, you try to work more in a range of amounts. The range for ale yeast is 375 million to 1 billion cells per liter. It depends on which yeast you use, the style of beer you are making and the flavor you are looking for.
Proper pitching rates for lagers is higher.
There are many books and internet forum posts on the subject.

No. The answer for all intents and purposes is no. Typically you are adding a few percent of the fermenting population, lag phase is the reproductive phase when yeast multiplies to critical fermentative mass. 5 lbs of yeast in a 5 gallon carboy is excessive but an extra 5 grams in a carboy doesn't make a blip on the logarithmic scale of yeast population.
