I only have one kettle but want to do two batches on the same day to cut down on brewing several times a month. If I use my 10 gal igloo cooler to have one batch mashing in the bag and do another in the kettle, will letting the one in cooler sit longer after removing the bag with grains affect that wort while brewing the other?
 
Enzymatic activity takes place in the fluid phase, so you could leave the bag in there after the hour mash. You may end up with a slightly more fermentable wort as the enzymes remain active for the time it remains in their temp range.

Alternatively, you could start the mash of the 2nd batch right after you've lautered the wort or start the boil of the first. For that you may need to temporarily store your lautered wort from the 1st batch so you can fill the mash tun with strike water for the 2nd batch.

It depends on how "quickly" is interpreted. That's why we do a mash out when fly sparging for an hour, to be sure, isn't it?
