1) With WhiteLabs yeast, do I need to let the yeast in the vial sit out for the 3-6 hours prior to putting it into the starter glass?

2) How long can I leave the starter out for? If I make it tonight and plan on brewing about mid-day on Saturday (so about 1.5 - 2 days) will it be ok?
 
yes and yes

Edit:
If you have enough time (which you do) I would give your starter 24-48hr then crash cool it and decant off the starter beer before pitching.

After you cool it in the fridge, the yeast will settle tightly on the bottom of your flask. You can dump out most of the liquid solution on top and leave a small amount to rouse up the yeast before pitching.

Decant means to pour of the liquid. This is the method I use for large starters so you don't dump all that "junk beer" into your brew. Let it ferment out completely, then put it in the fridge overnight to let the yeast settle to the bottom, then carefully pour off the liquid. You can leave just enough liquid to help swirl up the yeast when you go to pitch.
