I saved 400ml of washed yeast sludge for 2 weeks in the refrigerator and marked the sludge level on the flask. I decanted and added 1.5 l wort to make a starter that ran for 1 1/2 days on a stir plate. After cold crashing I noticed no increase in sludge level. My idea was with the increase in sludge I can calculate the growth rate and with that the cell count of my original 400 ml.
Is it possible that the concentration of the harvested yeast was so high that the growth was zero?
The yeast was alive because after pitching, a healthy fermentation started hours later. (WY3787)
