I am expecting my Samual Adams Summer Ale kit tomarrow to brew on Saturday and had a question about the Yeast Nutrient that comes with it.

Is this to be used to in the starter, instead of the starter, in the primary, or in conjunction with DME in the starter???

The instructions say to use it in the boil. I assume you could use it in the starter, but I can not say for sure, I would only be guessing, you have to ask them to make sure.
 
If you're referring to what I think, a little pill-like capsule, then you add it to the last 10 minutes of the boil.
 
the Wyeast nutrient I have (a big plastic vial/tube thing) says to mix in to the wort 10 mins before the end.

i'd do it in the primary wort, not the starter wort.
