I take my first just before I pitch my yeast and not again till I either transfer to secondary (two weeks) or if no secondary just before I keg (usually after three weeks).
 
Take one right before you pitch your yeast. Take another in a couple of weeks. Take a third a couple of days later. If the last two agree, fermentation is done (though you might wants to give it a little longer to clean up and age).
 
Ideally, never during fermentation, but before and after. I'm not in such a hurry that I need to know the day fermentation stops. Long primaries have proven to make my beers better, so most of the time my beer has been done fermenting for a week or two by the time I check the FG.
