digging up hops

I don't like the spot I chose 2 years ago. The amount of light changes drastically later in the season. Can I just dig up the roots and replant? Can I chop them up and make multiple new plants?

Yes and yes! I'm not a fan of my spot this year either and was planning on doing the same as you in a couple of months.

I transplanted a couple of plants last year. Simple as a shovel and a little elbow grease.

yes, though they will probably still grow in the old area. You can keep digging up the extra shoots.
