Reusing yeast and calculating pitch count

When you reuse yeast and by washing it and storing it in 4 pint jars, is it then possible to accurately calculate the amount of starter needed for a batch using one of those jars of yeast? On the pitch count calculator, you have to know how many cells of yeast you are starting with. Do people just guestimate that 1 jar has about 100 billion cells in it?
 
not terribly accurately, but the mr. malty calculator has a slurry tab that should help. If you want better than that, you'll have to get a haemocytometer and a scope.
