I am assuming you're using an 11 g packet? I would personally be too scared to try to save it and risk contamination. For 3 bucks its worth the piece of mind to just get another packet later.

If its for a block of dried yeast I'd sanitize a canning jar and vacuum seal it.
 
I have split US-05 before when I was doing 2.5 gal batches. I clip the corner of the yeast packet & weigh out 5.75 g in a sterilized container. Roll the packet up & tape tight, then place in a ziplock bag in the refrigerator. I have stored yeast this way for a month with no problems.
 
For 2.5 gallon's I just put the whole 11 gr in.

When I take the envelope out of the refrigerator I put it in my pocket to warm up before pitching. Never had a any trouble getting it to start fermenting with in six hours.
