conical yeast harvesting

So I've been using my mini-brew conical for a year and have never tried to harvest the yeast. So I figured I would give it a run. I've read several of the pages about washing the yeast, but have on question. When should I take it from the conical? I've been pulling trub off every few days. Should I just let the trub build up for several days and wash it? Or, after I keg, do I leave a bit in the fermenter and then drain the sludge and remainder of the liquid and harvest the yeast from this? Thanks
 
I take a few pulls in the first couple days to get most of the trub out. I like to take my yeast pull during the middle of fermentation. You don't want the yeast that floc's out to early or near the end. So for me about half way I get me my sample and use that for storing or repitching.
