I am going to attempt my first starter with white labs east coast ale yeast. Do I take it out of the fridge 3 to 6 hours like I normally would if I were direct pitching or do I leave it in the fridge until I am ready to pitch it into my starter growler?

I do. In general you always want temp changes for the yeast to be gradual, throwing them into 70 or 80 degree water from the fridge is not a good idea.
