I am thinking of using the yeast cake from my Pliny DIPA batch and pitch it into a big RIS batch. Will this work. I used US-56 for the Pliny.

What's the best way to harvest this yeast cake for re-use?
 
Those yeast are probably kind of tired, you might want to brew a low OG session beer on the cake before taxing it with a RIS, just to get the yeast refreshed and ready to go.
