I'm using a blow off tube for the first time and have concerns that it's not making a proper seal. It has been 12 hours since I pitched the yeast and I'm not seeing any activity. Any advice would be appreciated.
 
Wouldn't worry much after only 12 hours. It often takes a full day or even 2 or 3 to show signs of fermentation.
 
Also keep in mind that it'll take a bit more pressure to make them big ol' blow off bubbles. Its not like blowing through a tsp of liquid in an airlock.

Give it time, you're probably fine.
 
Advice? There is a sticky above this very topic that warns you about fermentation taking up to 72 hours to shows visible signs.

Relax.
