Picked up a used 20lb tank and regulator - To add to my kegging starter kit of two 10lb tanks (one backup for instant replacement of spent one) and regulator.

Should I carb from the 20lb and serve from the 5lb, or vice versa?? I have two deep chest freezers that are on temp control.
 
That's pretty much what I do. I have a chest freezer in my garage with a 20lb tank and a kegerator upstairs with a 5lb tank. All kegs get carbed in the chest freezer and then moved upstairs and served on the kegerator.
 
For typical beer carbonated to 2.3 volumes, it takes about as much CO2 to carbonate 5 gallons as it does to push out 5 gallons at 12 PSI. In essence, it doesn't really matter which tank you use for each purpose since they will both empty out about the same amount.
