I am turning a 55 gallon plastic barrel into a mash tun with a full size false bottom. The top of the barrel has two plugs, one big and one little. I'm thinking I can flip the barrel over and use the little plug as the outlet for the mash tun. This hole isn't in the middle, is that going to cause me problems?

Shouldn't cause too much dead space. I suppose maybe the barrel lid would bow a bit under the weight causing your drain to be slightly off center. I guess the only way to tell is to test it out 
You might be able to run some sort of sideways diptube, but if you know you're going to lose 1/2qt to dead space, just mash in with that much more.

I see no problems there. The full size false bottom will allow flow though all parts of the grain bed. Drain location shouldn't matter.
