sanitizing tubing/white labs yeast?

First question. Tonight I bottled and was going to use the carboy cap that allows you to blow into it to start a siphon. I got my racking cane and tubing filled with starsan at which point I realized the carboy cap wasnt all that neccessary because I was ready to start a siphon. Seeing as how people find these caps usefull how are you sanitizing the inside of your tubing?

With the white labs yeast before it is pitched should you pour off the liquid? Isn't it beer that sits on top of the yeast?

Side note:Since I didn't have to blow into the carboy cap I forgot to take the little cap off when I started bottling. Im really impressed how much I was able to collapse the better bottle before I realized the flow rate was really slow.
