Those cone-shaped white ones are designed to fit between the male and female flare connections in your system. Some flare parts might have a washer built into them (the ball-lock disconnects usually do have some black plastic at the end of the threaded part), others you will need to use one of these washers to get it to seal.

The flat nickle sized washer goes between the CO2 Tank Out and the Regulator. It smashes in there to get a nice tight seal between the tank and the regulator.

I don't have a nickel sized washer. I have a small cone shaped white washer that was taped to the regulator.
 
The easy rule of thumb I was told is that if the connections are going to be metal-on-metal use one of those washers.
 
It is very possible that all of your connections already have the washers built in.

If that extra washer from the regulator is the same size/shape as the picture you posted, then it could be for the connection of the gas line to the regulator.
