    I'd use an undrilled rubber stopper. Same thing folks use to fit and air lock on a glass carboy, only without the hole. You can buy 'em undrilled. That way you can remove for cleaning.

    How do I determine how much DME for a starter

    The ratio I always use is 1 gram of DME for every 10 grams of water. Grams and mL are roughly equivalent for water at the temperatures we use. Easy to scale up with this ratio in mind.
