so assuming 100 lbs of nitrogen per acre per year, if I have fertilizer that is 25% nitrogen, I will need 400 pounds total per acre ?? ( I rounded the numbers to make easier)

Correct. 100# N/A/year, is this based off of a soil test?
 
Do a soil test for sure the ground is different everywhere


You should never do a soil test for basing your nitrogen applications. A soil test would be useful for baseline P and K, as well as any micronutrients needed, but N is so mobile that it will change between the time you collect and the time you apply that your application rate should be based on crop needs and not a soil test.
