Pitching rate question (Mr. Malty)

Background, I harvested some Heady Topper yeast and stepped it up to the 2.5 gallon mark so far. I'm about to harvest into separate mason jars for re pitching in later 5 gallon batches.

My question is, when re-pitching Mr. Malty says to use 124ml's of yeast. Is this 124ml from the solid yeast layer after settling (right side of picture) or is it based off the jar being agitated (left side)?

FWIW I might try checking with the brewery to see if they use the same yeast for bottling as they do for primary. Many do and many don't. Bottling yeasts usually don't taste very good though...
