Is there supposed to be a nylon washer on the connection to the CO2 regulator (not at the connection to the tank)? I bought a dual regulator setup 2 years ago and one of the regulators crapped out on me. Those two regulators have small nylon washers in the recessed portion of the regulator, but my replacement regulator that I got from kegconnection does not. I feel like it might just be tapered threads that seal with some teflon tape, but I just want to be certain.

The connection where the CGA nipple screws into the regulator body should just be NPT, so you are correct. It's a tapered thread that needs teflon tape. Keep in mind that some regulators (Taprite) use left-handed threads on these fittings. They're usually marked but not always.
