First time kegger...read the directions on my regulator...said to leave the cylinder valve all the way open (said its because the cylinder valve seals in two places) On other stuff I've read it says to just leave it open about 1/2 turn. Does it matter?

There was a rather lengthy discussion last spring. The valve on your tank has to be opened all the way and back-seated, or it will leak.

Yes it does matter. The valve on the CO2 cylinder has a front seat (seals when closed), and a back seat (seals when open). When you open the valve it needs to be opened all the way and just snugged up on the back seat.

I had never heard of this. Maybe that is why my first tank ran out so fast even though I could find no leaks.
