These methods just seem to be a simple starter. I want to start with a vial of 100 billion cells and growth it to 300-350 billoin cells. I need to learn how to step it up over several days and adding new wort to it.
Adding any thing to a culture will just lead to contamination, so its best to be avoided. on Ebay in the industrial/ lab supply section you can find great deals on large flasks, stirrers ect. if you want to get a big starter going use a large volume flask and twice to three times the nutrients in the media. That how we do it in the lab I work in.
I have a 1L cell culture flask that has a stir paddle in the lid, and I've been putting 1/2Cup of DME in to one liter and pitching a Wight labs flask into that the night before, then I tale 333ml and pitch that into a 6gal batch, the put the other 666ml
into two sterilized bottles and store them in the fridge for later brewing.
