Does anyone know how one would go about isolating individual strains of yeast from a wild culture?

"Streak plate."

You'd need a petri dish, malt agar, and inoculating loop... using sterile techniques.

This shows the technique.

Yep. Fairly straightforward technique as Mensa says. You can even do it as a homebrewer with minimal equipment and a bit of DIY effort.

Although it is worth noting that mere homebrewers will have a tough time identifying many of the different strains of yeast once they do have isolated cultures growing on slants/plates.

You're right! Let me just add to that equipment list:

a good microscope with at least 400X.
