I use a sterilized kitchen wisk for probably 30 seconds to a minute. I try to stir the wort pretty good so it gets full of bubbles and mixed around pretty well.
 
If you use pure oxygen, you'll only need a few minutes.

If you are aerating with a normal aquarium style aeration system, usually I aerate for about 20-40min. but I've heard of others aerating up to an hour. Prepare for a foamy time.
 
I use a diffusion stone with O2. I used to aerate 5 gallons for 1 minute, but started to detect some hot alcohol tastes, so I dropped down to about 30 seconds.
