Is it okay to make a large starter colony (milk jug) then transfer the yeast into sanitized beer bottles and put a cap on them? Can I then store them in the fridge and pull one out when ever needed?

I would put them under sterilized, distilled water instead of leaving them under the wort. That is if I was going to do what you are planning on doing. And, yes your plan will work.

Thanks for your response.

Would storing them on distilled water cause the cells to pop due to osmosis?

I guess storing them on sugar could cause them to shrink too...

There's an interesting post about pressure cooking water and put a little bit of yeast that can stored for a year plus. Might want to try finding it. Sorry don't have the link.
