I have one of those little O2 bottles from HD or Lowes. What's the best way to rig it up with hardware store pieces to oxygenate my wort .

Aquarium hose to brass transition to vinyl hose to stone barb.
I prefered to use more of the aquarium hose than the larger vinyl based soley on the aquarium hose wastes less volume after the valve is turned off.

Ooops. Assumed you had the regulator already.
There are torche kits that have the regulator valves in them but, for a quarter of that kit cost you can get a regulator from an online HBS.
