This is my first time using this type of yeast and I was wondering if you can take it right out of the fridge and activate than wait the 3-4 hours or if you need to let it get to room temperature before activating.

Leave it in the fridge until you activate it. I take mine out in the morning before brewing and just leave it on the counter and let it gradually reach room temperature after it's been activated.

You can also take it out even earlier if you want (6-8hrs)...so far I've had good results with Wyeast, fermentation started almost immediately after pitching the stuff on my last batch.
