If you're talking about brew systems, it's the number of stands (tiers) for kettles to sit on. Usually consists of one tier with a gas burner (for boiling) and another to collect the finished wort using gravity to transfer. Most folks, from what I know, use the 3 tier to sparge on the highest tier first, then transfer to boil, and so on.

If you mean the 3-tier distribution system, that's a whole other can o' worms.

Tiers are levels. A three tier setup utilizes gravity to move liquor or sweet wort from one vessel to the other. The less tiers, the more gadgets i.e., pumps needed to move the liquids.

Yes, barrel. A sanke 15.5 gallon keg is a half barrel.
A 3BBL is a 3 barrel system = 6 sankes.

3 Tiers means there are 3 vertical levels to the brewing rig. A single tier system is basically only a table and the HLT, MLT, and BK all sit next to each other. It's easier to get access to the equipment, but it requires pumps to move the liquids, and takes up more horizontal space than a vertical system (usually).
